    Aggregates of diketopyrrolopyrrole dimers in solution

    Dimers based on diketopyrrolopyrrole (DPP) chromophores have gained tremendous interest as an excellent material building block for organic solar cells and photodiodes. However, a counterintuitive blue-shift in their solution absorption spectra occurs with an increasing number of thiophene units bridging the DPP moieties. We allocate this to aggregates in solution, which might hinder adequate mixing in blends, leading to poor film forming quality and reduced charge generation in solution processed devices. Hence, identification of such aggregates is crucial in order to find measures for device optimisation. In this study, we present synthesis and characterisation of three pyridyl end-capped DPP dimers of different conjugation length using thiophene linkers and compare their parent monomer to evidence the behaviours of aggregates in solution. We employ conventional and temperature dependent UV–Vis spectroscopy, fluorescence and excitation-emission spectroscopy as well as TD-DFT calculations to show that such DPP dimers predominantly form aggregates in solution even at low concentrations. By disentangling the spectroscopic behaviour of both aggregated and non-aggregated species, we refute literature's explanation that the apparent blue shift in absorption arises from a reduced conjugation length due to more molecular flexibility. Instead, absorption and emission signals of non-aggregated dimers are mostly masked by their aggregated species. This work provides a tool set using common laboratory spectroscopic equipment to identify and characterise solution aggregates—information particularly important towards optimisation of organic electronics processed from solution

    Electrochemically tuneable multi-colour electrochemiluminescence using a single emitter

    A single starting component electrochemiluminescence system from which red, green, blue or white emission can be obtained, depending on the applied potential or the mode of the ECL experiment, is described. The convoluted ECL spectral responses observed at different potentials are readily explained using a 3D-ECL technique, where the ECL spectral profile is continuously monitored as a function of potential during voltammetric scanning. The 3D plots obtained using this technique implicate cross-annihilation ECL reactions involving the complex itself and stable products resulting from its electrolysis. Combining this information with knowledge of the energetic requirements of the various reactions involved, suggests a mechanism involving traces of two emissive products, related to the loss of a methyl group from the triazole moiety. These products, while barely detectable electrochemically, are sufficiently emissive to influence and even dominate the ECL emission under some conditions

    Complete genomic sequence of the temperate bacteriophage ΦAT3 isolated from Lactobacillus casei ATCC 393

    AbstractThe complete genomic sequence of a temperate bacteriophage ΦAT3 isolated from Lactobacillus (Lb.) casei ATCC 393 is reported. The phage consists of a linear DNA genome of 39,166 bp, an isometric head of 53 nm in diameter, and a flexible, noncontractile tail of approximately 200 nm in length. The number of potential open reading frames on the phage genome is 53. There are 15 unpaired nucleotides at both 5′ ends of the ΦAT3 genome, indicating that the phage uses a cos-site for DNA packaging. The ΦAT3 genome was grouped into five distinct functional clusters: DNA packaging, morphogenesis, lysis, lysogenic/lytic switch, and replication. The amino acid sequences at the NH2-termini of some major proteins were determined. An in vivo integration assay for the ΦAT3 integrase (Int) protein in several lactobacilli was conducted by constructing an integration vector including ΦAT3 int and the attP (int-attP) region. It was found that ΦAT3 integrated at the tRNAArg gene locus of Lactobacillus rhamnosus HN 001, similar to that observed in its native host, Lb. casei ATCC 393

    Nano-Alignment in Semiconducting Polymer Films: A Path to Achieve High Current Density and Brightness in Organic Light Emitting Transistors

    Organic light emitting field effect transistors (LEFETs) integrate light emission of a diode with logic functions of a transistor into a single device architecture. This integration has the potential to provide simplified displays at low costs and access to injection lasing. However, the charge carrier mobility in LEFETs is a limiting factor in realizing high current densities along with a trade-off between brightness and efficiency. Herein, we present a technique controlling the nanoscale morphology of semiconducting polymers using nanoscale grooved substrates and dip-coating deposition to achieve high current density. We then applied this approach to heterostructure LEFETs and demonstrated brightness exceeding 29000 cd m–2 at an EQE of 0.4% for a yellow emitter and 9600 cd m–2 at an EQE of 0.7% for a blue emitter. These results represent a significant advancement in organic optoelectronics and are an important milestone toward the realization of new applications in displays and electrically pumped lasing

    Increased ATP generation in the host cell is required for efficient vaccinia virus production

    To search for cellular genes up-regulated by vaccinia virus (VV) infection, differential display-reverse transcription-polymerase chain reaction (ddRT-PCR) assays were used to examine the expression of mRNAs from mock-infected and VV-infected HeLa cells. Two mitochondrial genes for proteins that are part of the electron transport chain that generates ATP, ND4 and CO II, were up-regulated after VV infection. Up-regulation of ND4 level by VV infection was confirmed by Western blotting analysis. Up-regulation of ND4 was reduced by the MAPK inhibitor, apigenin, which has been demonstrated elsewhere to inhibit VV replication. The induction of ND4 expression occurred after viral DNA replication since ara C, an inhibitor of poxviral DNA replication, could block this induction. ATP production was increased in the host cells after VV infection. Moreover, 4.5 μM oligomycin, an inhibitor of ATP production, reduced the ATP level 13 hr after virus infection to that of mock-infected cells and inhibited viral protein expression and virus production, suggesting that increased ATP production is required for efficient VV production. Our results further suggest that induction of ND4 expression is through a Bcl-2 independent pathway

    Activation of β-Adrenoceptors by Dobutamine May Induce a Higher Expression of Peroxisome Proliferator-Activated Receptors δ (PPARδ) in Neonatal Rat Cardiomyocytes

    Recent evidence showed the role of peroxisome proliferator-activated receptors (PPARs) in cardiac function. Cardiac contraction induced by various agents is critical in restoring the activity of peroxisome proliferator-activated receptors δ (PPARδ) in cardiac myopathy. Because dobutamine is an agent widely used to treat heart failure in emergency setting, this study is aimed to investigate the change of PPARδ in response to dobutamine. Neonatal rat cardiomyocytes were used to examine the effects of dobutamine on PPARδ expression levels and cardiac troponin I (cTnI) phosphorylation via Western blotting analysis. We show that treatment with dobutamine increased PPARδ expression and cTnI phosphorylation in a time- and dose-dependent manner in neonatal rat cardiomyocytes. These increases were blocked by the antagonist of β1-adrenoceptors. Also, the action of dobutamine was related to the increase of calcium ions and diminished by chelating intracellular calcium. Additionally, dobutamine-induced action was reduced by the inhibition of downstream messengers involved in this calcium-related pathway. Moreover, deletion of PPARδ using siRNA generated the reduction of cTnI phosphorylation in cardiomyocytes treated with dobutamine. Thus, we concluded that PPARδ is increased by dobutamine in cardiac cells

    Increasing utilization of Internet-based resources following efforts to promote evidence-based medicine: a national study in Taiwan

    BACKGROUND: Since the beginning of 2007, the National Health Research Institutes has been promoting the dissemination of evidence-based medicine (EBM). The current study examined longitudinal trends of behaviors in how hospital-based physicians and nurses have searched for medical information during the spread of EBM. METHODS: Cross-sectional postal questionnaire surveys were conducted in nationally representative regional hospitals of Taiwan thrice in 2007, 2009, and 2011. Demographic data were gathered concerning gender, age, working experience, teaching appointment, academic degree, and administrative position. Linear and logistic regression models were used to examine predictors and changes over time. RESULTS: Data from physicians and nurses were collected in 2007 (n = 1156), 2009 (n = 2975), and 2011 (n = 3999). There were significant increases in the use of four Internet-based resources – Web portals, online databases, electronic journals, and electronic books – across the three survey years among physicians and nurses (p < 0.001). Access to textbooks and printed journals, however, did not change over the 4-year study period. In addition, there were significant relationships between the usage of Internet-based resources and users’ characteristics. Age and faculty position were important predictors in relation to the usage among physicians and nurses, while academic degree served as a critical factor among nurses only. CONCLUSIONS: Physicians and nurses used a variety of sources to look for medical information. There was a steady increase in use of Internet-based resources during the diffusion period of EBM. The findings highlight the importance of the Internet as a prominent source of medical information for main healthcare professionals

    Mechanical Characterization of Rabbit Pulmonary Vein Sleeves in In Vitro Intact Ring Preparation

    BackgroundPulmonary vein (PV) sleeves, composed of cardiomyocytes, play certain roles in arrhythmogenesis. In the literature, it has been frequently reported that PV sleeves possess intrinsic spontaneous pacemaking activity and triggered activity in normal dogs and rabbits. In contrast, other research groups presented totally opposite findings which showed absence of such pacemakers in dogs, rabbits and rats. The present study was designed to clarify this puzzle and contradiction.MethodsA novel methodology using in vitro experimentation was used to examine the electromechanical activity of whole segments of PV sleeves. The ring preparation was composed of a small piece of left atrial (LA) free wall, PV ostium and sleeve from rabbits. A circumferential contraction of the PV sleeve was measured when the preparation was electrically driven from the LA free wall. Mechanical force of the ring preparation was measured using a force transducer. The action potentials were recorded using conventional intracellular recording technique in strip preparation.ResultsIn 15 rabbits, no spontaneous pacemaking activity or triggered activity was found in the in vitro ring preparation of PV sleeve. The circumferential contraction of PV sleeves was external calcium-dependent. Frequency-force relation displayed a negative staircase at 0.1–0.5 Hz and a positive staircase at 1–5 Hz. Post-rest potentiation was prominent between 15 s and 120 s. Intracellular action potential recording did not display any automaticity or triggered activity in PV sleeves.ConclusionIn an intact ring preparation of rabbit PV sleeves, intrinsic spontaneous pacemaking activity or triggered activity was not found
